STAT3報(bào)告基因是自主研發(fā)的用于檢測(cè)STAT3二聚體和STAT1/STAT3異質(zhì)二聚體的轉(zhuǎn)錄活性水平為目的的報(bào)告基因。激活STAT3后形成STAT3二聚體和STAT1/STAT3異質(zhì)二聚體，它們與SIS-inducible element（SIE）反應(yīng)元件進(jìn)行相互作用調(diào)控細(xì)胞的生長(zhǎng)和凋亡。STAT3(signal transducers and activators of transduction-3)報(bào)告基因與多種惡性腫瘤的發(fā)生、發(fā)展和侵襲轉(zhuǎn)移關(guān)系密切。
STAT3報(bào)告基因主要用于監(jiān)控細(xì)胞STAT3信號(hào)通路中的STAT3轉(zhuǎn)錄活性、腫瘤****的研發(fā)以及基因過(guò)表達(dá)和RNAi的表型分析等。
pGMSTAT3-GFP是改造后的哺乳動(dòng)物真核表達(dá)載體，在其多克隆位點(diǎn)插入了多個(gè)STAT3結(jié)合位點(diǎn)，可以高效地檢測(cè)STAT3的激活水平。由于載體采用了GFP作為報(bào)告基因，更便于后續(xù)的檢測(cè)。同時(shí)，對(duì)載體中預(yù)測(cè)出的其它轉(zhuǎn)錄因子以外的結(jié)合位點(diǎn)進(jìn)行了適當(dāng)?shù)耐蛔?，增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。另外，由于質(zhì)粒體積減小，使得STAT3-GFP報(bào)告基因更易于轉(zhuǎn)染。STAT3 GFP Reporter Plasmid（STAT3-GFP報(bào)告基因質(zhì)粒）

pGMSTAT3-GFP可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動(dòng)物細(xì)胞。
